The win moves the Welsh club back to second in the table, a day after being overtaken by Yeovil, and back to within five points of table-topping Scunthorpe.
Marc Goodfellow scored the winner, heading home after 61 minutes.
The goal came moments after Oxford had been reduced to ten-man, when Jon Ashton was shown a second yellow card.
With United playing the Swans at Glanford Park on Saturday, it means a home win would extend the gap at the top of the table to eight points.
A win for visitors, however, would cut the gap to just two points.
