With the game being the first meeting between the sides in 14 years, the police are taking no chances and will have a large number of officers at the scene.
Officers will patrol Glanford Park, as well as the town centre and train station in a bid to head of any potential trouble.
With more than 8,000 fans heading to the big match, it only takes a small minority of troublemakers to spoilt it for the majority.
Humberside police are also mindful of a 'meet' in April between rival Scunthorpe and Cod fans.
A town centre brawl, involving Cod 'fans' returning from a League fixture at Stockport and Iron 'fans' is the worst ever football violence seen in North Lincolnshire. A series of raids last month led to more than 40 arrests in Scunthorpe and Grimsby.
Sargent Brian Burns said: "The police presence will be strong, with staffing increased due to intelligence and the potential for disorder.
"There will be a substantial police presence, both in and around the ground as well as in the town centre before and after the mathc with uniformed officers being supported by dog handlers, mounted section and evidence gathering teams.
"This is a game of interest for supporters on both sides, in view of this licensees have been advised of the fixture and a large proportion of pubs in the town have stated they intend to close or restrict entry to regulars."
Scunthorpe United have also discovered forged tickets in circulation for the match, and fans without tickets - or with false tickets - have been told to stay away from the game."
Sargent Burns added: "The police and the club will be mounting an operation to check tickets, and anyone caught with one will be arrested."
