Hooper has a hamstring injury meaning Jonathan Forte will lead the line for the Iron, probably alongside Paul Hayes.
Nigel Adkins, the Scunthorpe manager, is also without Ben May (ankle) while Garry Thompson (foot) is pushing to be fit.
Port Vale have no fresh injuries as they look to bounce back from Saturday’s 1-0 home loss to Bury in League Two.
They are without the former Iron striker Geoff Horsfield (groin), James Lawrie (appendix) and goalkeeper Joe Anyon (broken leg).
