The semi-final at Glanford Park will be the Iron's third game of pre-season, but the first one on home soil after 1-0 wins over Bray Wanderers and Wayside Celtic during a training camp in the Republic of Ireland.
Centre-back Paul Reid (hand) and midfielder Damien Mozika (ankle) will sit out the game, while a number of trialists will help fill out the home squad.
That means Tom Newey, Matty Mainwaring and Yami – who all featured in Ireland - could get further run outs against the Mariners.
Grimsby are the defending champions, having beaten fellow Blue Square Premier side Lincoln 4-3 in last year's final.
The Iron had dominated the competition prior to that, lifting the trophy for four consecutive seasons prior to a semi-final loss at Lincoln last pre-season.
Striker Liam Hearn (Achilles) will be missing for the Mariners.
Only the Scunthorpe Telegraph Stand will be open for Iron fans on Tuesday night, with entrance £6 for adults and £3 concessions. The Iron Bar will remain closed, but the Old Showground Bar in the stand will be open.
