Centre-half Mirfin missed the home defeat to Accrington at the weekend due to a minor calf injury but should be fit to partner Niall Canavan at the heart of defence.
Terry Hawkridge is also likely to come into the side after recovering from illness.
The Iron will be without new loan signing Reuben Noble-Lazarus, who joined from Barnsley last week in time to make his debut off the bench on Saturday, but is ineligible having not been at the club when the original tie took place.
Defender Eddie Nolan had been hoping to return at the weekend, but is still struggling with a hamstring injury and midfielder Matt Sparrow is still out with a similar problem.
Grimsby will check on Liam Hearn and Ritchie Jones.
Forward Hearn missed the 2-0 league win at Tamworth at the weekend with an ankle injury while midfielder Jones has been absent through illness since the initial tie at Blundell Park, which finished goalless, nine days ago.
Chris Doig will also continue to be monitored but Andy Cook, Alex Rodman and Paddy McLaughlin all returned to the squad on Saturday and came on as substitutes.

Iron boss Brian Laws insists there has been a positive response in training after the 2-0 defeat against Accrington on Saturday.
“We had a really good meeting on Monday morning, which was important after the weekend,” Laws told the club's official website.
“We had to identify how and why it went wrong and we’ve done that.
"There has been a positive response from the players and there is no stone unturned from Saturday’s game. What we are looking for is for a response and an improvement from what we put out there on Saturday.”

Codfather Paul Hurst is in a buoyant mood following their 2-0 win at Tamworth and told the Grimsby Telegraph: "If they go out tonight and give me everything – which they have been doing, then, whatever the result, I'll still be proud of them.

When things are rushed, they usually go tits up and that's argubally the case with tonight's man in the middle Stuart Attwell. Demoted quicker than he was fast tracked to the Premier League, the giver of the famous ghost goal at Watford is averaging just three yellow cards a game this year- he must have calmed down!
Attwell was berated by the majority of gaffers and argued to be the 'worst referee in the history of the Premier League' during his short stint, before being summoned back to the Football League by angry ref inspector Mike Riley a few years ago. Let's hope he doesn't spoil it tonight.

No tickets are available after 4pm on the day of the game on police advice. Less than 4,000 tickets have been sold for the home areas of Glanford Park.
