The Iron are seeded in the northern section of the draw, meaning they will face one of Carlisle, Oldham, Tranmere, Hartlepool, Rochdale, Stockport, Morecambe, Rotherham, Chesterfield, Bury, Bradford, Accrington Stanley, Crewe, Macclesfield or Lincoln.
Nottingham Forest, Leicester, Sheffield United, Middlesbrough, Doncaster, Derby County, Preston, Barnsley, Coventry, Leeds, Sheffield Wednesday, Peterborough, Huddersfield and Walsall have also been seeded so cannot be drawn against Scunthorpe.
The draw will be released at 9.30am. United will also learn their Championship fixture list on Thursday.
